چکیده

Biomimetic scaffolds are desirable for 3D cell culture, yet optically transparent, macroporous, polysaccharide hydrogels have been limited. By incorporating soluble monoand disaccharide additives during cryogelation, we synthesized biomimetic scaffolds and gained insight into the mechanism of their formation. The mono/disaccharide additives affect the size of ice crystal porogens and interact with polysaccharide polymers, altering cryogel pore size and mechanical properties. Importantly, gel transparency is maintained following removal of additives, enabling their use as biomimetic extracellular matrices. We demonstrate both optical transparency with 3D spatial control of immobilized bioactive growth factors using multiphoton patterning and cellular response to immobilized ligands.

Bio-inspired fabrication of fibroin cryogels from the muga silkworm Antheraea assamensis for liver tissue engineering.

Conventional scaffold fabrication techniques result in narrow pore architectures causing a limited interconnectivity and use of porogens, which affects the bio- or cyto-compatibility. To ameliorate this, cryogels are immensely explored due to their macro-porous nature, ease in fabrication, using ice crystals as porogens, the shape property, easy reproducibility and cost-effective fabrication te...

Fabrication of Biomimetic Bone Tissue Using Mesenchymal Stem Cell-Derived Three-Dimensional Constructs Incorporating Endothelial Cells

The development of technologies to promote vascularization of engineered tissue would drive major developments in tissue engineering and regenerative medicine. Recently, we succeeded in fabricating three-dimensional (3D) cell constructs composed of mesenchymal stem cells (MSCs). However, the majority of cells within the constructs underwent necrosis due to a lack of nutrients and oxygen. We hyp...

Tissue mimetics: engineered hydrogel matrices provide biomimetic environments for cell growth.

T issue-engineered scaffolds were initially designed for implantation with the idea of growing tissues in vitro before transplantation. This spawned several decades of research, which is still active today, into highly porous polymeric scaffolds in which cells are seeded and expected to develop into tissue. Biomaterial matrices were designed to provide structured extracellular matrix (ECM) anal...

Highly porous chemically modified carbon cryogels and their coherent nanocomposites for energy applications

Highly porous carbon cryogels with tunable pore structure and chemical composition were synthesized through controlled hydrolysis and polycondensation reactions using different chemicals as precursors and either NaOH or hexamine (C6N12N4) as catalysts. Gelation was followed with freeze drying to preserve the highly porous structure during solvent removal and controlled pyrolysis of the organic ...

Cartilage-like electrostatic stiffening of responsive cryogel scaffolds

Cartilage is a structural tissue with unique mechanical properties deriving from its electrically-charged porous structure. Traditional three-dimensional environments for the culture of cells fail to display the complex physical response displayed by the natural tissue. In this work, the reproduction of the charged environment found in cartilage is achieved using polyelectrolyte hydrogels based...
